Abstract

Work to be Done in Final Year at UCSB

  1. We will begin the final amplicon work of the invertebrates using a COI amplicon. All 1000 samples will be used for analysis. These amplicons will be sequenced at OSU on the same machine we previously used to maintain methodology uniformity.
  2. We are completing the extractions and PCR for the year 4 of sampling. These samples will generate 2 sets of amplicon data for the microbes and the fish analysis.
  3. In this last year the Vega Thurber lab will a) extract DNA from all remaining time points, b) conduct amplicon generation for 16S (bacteria), ITS2 MiFish (fish), and COI (invertebrates) and then have them sequenced at OSU. These amplicons will be sequenced at OSU on the same machine we previously used to maintain methodology uniformity.
  4. We will ideally complete the 16S and fish analysis in this year. We suspect the analysis of the invertebrate data will continue past the end date.
